Elevated concentrations of serum creatine kinase are found in all types of muscular dystrophy and disease involving the destruction of muscle tissues.  In the case of progressive muscular dystrophy,  elevated CK concentrations may be present before clinical symptoms appear. Elevated CK concentrations are observed after myocardial infarction and in cases of acute cardiovascular disease and Reyes Syndrome.  Neurogenic musclediseases such as multiple sclerosis and polio do not produce elevated CK concentrations.  CK activity is in inverse relation to thyroid activity, thus, in cases of hypothyroidism, elevated concentrations of CK would be observed.

The Clinisciences enzymatic CK assay is a liquid stable 2-part system which is intended for in vitro quantitative determination of total creatine kinase in human serum and plasma. In the assay, creatine phosphate and adenosine diphosphate (ADP) are use as substrates, N-acetylcysteine (NAC) is the thiol activator, and adenosine monophosphate (AMP) and Diadenosine pentaphosphate (Ap5A) are added to inhibit interference caused by adenylate kinase activitony.

The rate of increase in absorbance at 340 nm due to the formation of NADPH is directly proportional to the creatine kinase activity.
